Summary
Trypanothione is a spermidine-glutathione conjugate unique to trypanosomatid parasites (Leishmania spp., Trypanosoma brucei, T. cruzi), where it functions as the central thiol-redox metabolite. Because it is absent in mammalian cells and essential for parasite survival, both trypanothione itself and the enzymes of its biosynthesis and recycling (trypanothione reductase, trypanothione synthetase) are intensively studied as targets for antiparasitic drug discovery. It is not administered therapeutically but is a research biochemical and drug-design reference molecule.
Mechanism of Action
Trypanothione [N1,N8-bis(glutathionyl)spermidine] is a unique dithiol molecule found in trypanosomatid parasites. It is the primary low-molecular-weight thiol in these organisms, maintained in its reduced form by trypanothione reductase (TryR). It serves as the main redox buffer replacing the glutathione/glutathione reductase system found in host cells, detoxifying reactive oxygen species, hydroperoxides, and xenobiotics. It is not itself a drug but rather an endogenous metabolite and validated drug target in Leishmania, Trypanosoma brucei, and Trypanosoma cruzi.
